Can I connect my DNS-323 straight to my computer with a Cat6 cable, bypassing a router?
Thanks in advance

Only if your NIC supports auto sensing.
If not you will need a crossover cable.

the DNS-323 nic is gigabit an therefor supports Auto MDI/MDI-X senseing itself. You can.

You only need to be able to cross once - you can cross in the cable, or you can cross in the hardware at one end, doesn't matter which.
BTW - I understand the Auto-MDIX feature is optional in gigabit - but I have yet to see a gigabit NIC that didn't support it.
